I was logistically prepared with a farm elsewhere but I expected these wolves to conquer or at least live long enough to reproduce.

Most weapons do at least 3 times the damage from behind, so I would venture a 'yes' here.

Also it seems like pets are much, much softer after being tamed, as if their armor were reduced or something. Maybe it just seems that way because they tend to get several enemy at once.

If only there were some way to buff pets so they were tougher. Hmm. ;)

Wolves are vulnerable to stagger (50% more than other creatures) and when staggered creatures (and the player) take double damage. The damage a deathsquito does is more than enough to stagger even a 2 star wolf.

Maybe the squito somehow managed to sneak attack the wolf for multiplied damage.

Wolves are pretty weak hp wise, guess that is made up with there bite, but I had one and a Dwarf Brute took it out with 2 hits

Deathsquitos have "death" in their name for a reason! Wolves have pretty low HP in general and I would recommend avoiding higher lvl biomes with your pets unless you are prepared to lose them.
