I think resistances always override any weakness. There’s a little bit of sketchiness when it comes to immunity and resistances though (serpents are resistant to fire because they are always wet, but technically are immune to it when not wet)

According to the wiki, any level of resistance always overides weakness, so yes, that deficiency of the root harnesk is easily surmountable once you get access to barley wine.

Also regarding the serpents; they just have a flat immunity to fire, it's not related to them being wet, they just ignore it outright. Though they are weak to frost, so better to use that on them instead.
