The safest, most conservative thing to do is a new seed. Just think on it as a "portal world" since you can of course move back and forth between seeds. Many community servers do this because 1 map is not enough and have resource seeds that they wipe every few weeks for those needing to get resources while 1 or 2 are main building/town servers.
For ease you know you can also use the same seed with a different name(clone if you will) it'll be new though so you'd have to move stuff from your 1st version to the second version. Naming your seed is what makes the seed unique to your ecocide if you will :P
If you haven't fully explored your current seed and have been -----no where----- near mistlands biomes on your map, then it probably is safe to assume that the mistlands update will spawn everything intended for your specific seed.
However if you've skirted mistlands areas but not revealed them on the maps fogged out area, there is a high chance not all parts of the mistlands biomes will spawn. This is intentional by the devs to better ensure that it doesn't disrupt/break someone's seed if they've done anything on it.
So all in all, I personally recommend just having multiple seeds to best ensure no discrepancies of getting or not getting the mistlands on the map properly when the update goes out.

A dev a few weeks ago said that the turn around on major updates is between 6 to 9 months. Since icecaves came out a couple of months back, you're looking at a mistlands release corridor most likely between september and december respectively. I'd say look to december and be pleasantly surprised if its before that. Given that so far we're primarily been seeing still concept art and some small feature pieces(troll traps this week were shown) with ingame stuff, the december end of the spectrum seems more compelling.
